I'm gonna use a anti fouling paint on the bottom. & 2pk top paint. applied by roller & brush Yes, you can (and should) use a barrier coat for a boat that stays in the water. Interlux Interprotect is probably the most commonly used product. The barrier coat is a good idea on the bottom (between your antifoulant and your gelcoat) to prevent water from permeating your gelcoat... if, in fact, your boat stays in the water. If you trailer the boat, don't bother. Just appropriately sand the bottom so the antifoulant will stick.
You don't really need to barrier coat your topsides. Just use a regular primer.
